Can the new 5S 4000mAh battery be used in place of a 4S 4000mAh battery with no changes to ESC or prop?

That depends on how close to the current rating of the ESC and motor the system is, on 4S. The motor will want to spin 25% faster, which is a fairly big increase in power. Also, you have to determine if the prop can safely turn at that rpm, as they have limits, too.
For example, my T28 Trojan peaked at ~30 amps on 3S with the stock prop. With a 4S battery, it went over 40 amps. The stock ESC was 30 amps, so I had to install a 45 amp ESC; the motor and prop were OK at the higher current and rpm. Now, I have a watt meter to verify all this, which every electric flyer should have, IMO.
